Cependant, il peut \u00eatre difficile de d\u00e9tecter le cancer \u00e0 ses premiers stades en raison de la biologie de la maladie et des limites des technologies actuelles. Une approche prometteuse consiste \u00e0 rechercher des biomarqueurs, comme les mol\u00e9cules d&rsquo;ADN et de petit ARN, qui sont sp\u00e9cifiques aux cellules canc\u00e9reuses. Cependant, la mesure de ces biomarqueurs est actuellement difficile et co\u00fbteuse, car elle n\u00e9cessite des \u00e9quipements et des laboratoires sp\u00e9ciaux.<\/p>\n<p>L&rsquo;ADN est la mol\u00e9cule qui transporte l&rsquo;information g\u00e9n\u00e9tique dans nos cellules, mais il peut aussi \u00eatre utilis\u00e9 pour cr\u00e9er des structures minuscules et pr\u00e9cises ayant des fonctions sp\u00e9cifiques. Ce processus, appel\u00e9 origami d&rsquo;ADN, s&rsquo;apparente \u00e0 l&rsquo;art japonais du pliage du papier. Des chercheurs de l&rsquo;<a href=\"https:\/\/www.unifr.ch\/home\/fr\/\" target=\"_blank\" rel=\"noopener\">Universit\u00e9 de Fribourg<\/a> utilisent cette technique pour cr\u00e9er des nanocapteurs d&rsquo;ADN capables de d\u00e9tecter des biomarqueurs du cancer, comme des mol\u00e9cules d&rsquo;ADN et de petits ARN, avec une pr\u00e9cision et une sensibilit\u00e9 \u00e9lev\u00e9es. Deux \u00e9quipes, dirig\u00e9es par le professeur Curzio R\u00fcegg (m\u00e9decine) et le professeur Guillermo Acuna (physique), travaillent ensemble sur ce projet dans le cadre du programme du <a href=\"https:\/\/www.snf.ch\/fr\/P72Gs36BEp0TlMoG\/page\/pointrecherche\/poles-de-recherche-nationaux\/materiaux-bio-inspires\" target=\"_blank\" rel=\"noopener\">programme \u00ab Mat\u00e9riaux bioinspir\u00e9s \u00bb de la PRN<\/a>.<\/p>\n<h4>Une nouvelle g\u00e9n\u00e9ration de biocapteurs<\/h4>\n<p>Ivana Domljanovic, doctorante \u00e0 l&rsquo;Universit\u00e9 de Fribourg, a cr\u00e9\u00e9 un biocapteur en origami d&rsquo;ADN en forme de livre qui peut d\u00e9tecter des petits ARN (miRNA) sp\u00e9cifiques li\u00e9s au cancer du sein. Ce biocapteur produit un fort signal fluorescent lorsque le miRNA est pr\u00e9sent et peut d\u00e9tecter deux miRNA diff\u00e9rents en l&rsquo;espace de dix minutes, \u00e0 un co\u00fbt tr\u00e8s faible. Cette recherche a \u00e9t\u00e9 r\u00e9cemment publi\u00e9e dans la revue <a href=\"https:\/\/www.rsc.org\/journals-books-databases\/about-journals\/nanoscale\/\" target=\"_blank\" rel=\"noopener\">Nanoscale<\/a>.<\/p>\n<p>Samet Kocabey, assistant principal et boursier Marie Curie-Sklodowska, a mis au point une plateforme en origami d&rsquo;ADN qui peut d\u00e9tecter jusqu&rsquo;\u00e0 quatre miARN diff\u00e9rents. Cette plateforme utilise un syst\u00e8me de code-barres g\u00e9om\u00e9trique et la technique de microscopie \u00e0 super-r\u00e9solution appel\u00e9e DNA-PAINT (Point Accumulation for Imaging in Nanoscale Topography) pour la d\u00e9tection. Cette m\u00e9thode est tr\u00e8s sensible, capable de d\u00e9tecter les miRNA \u00e0 des niveaux tr\u00e8s bas, et peut d\u00e9tecter sp\u00e9cifiquement les changements de base unique dans le miRNA. Cette recherche a r\u00e9cemment \u00e9t\u00e9 publi\u00e9e dans la revue <a href=\"https:\/\/www.sciencedirect.com\/journal\/biosensors-and-bioelectronics\" target=\"_blank\" rel=\"noopener\">Biosensors and Bioelectronics<\/a>.<\/p>\n<p>Les r\u00e9sultats de ces projets ont des implications importantes pour les recherches futures. Ils constituent une base pour la cr\u00e9ation de capteurs plus avanc\u00e9s, capables de d\u00e9tecter plusieurs types d&rsquo;acides nucl\u00e9iques en m\u00eame temps (multiplexage), ou de d\u00e9tecter des changements dans les mol\u00e9cules d&rsquo;ADN ou d&rsquo;ARN (ARNm). Cela peut \u00e9galement conduire au d\u00e9veloppement de tests de diagnostic du cancer plus rapides, plus s\u00fbrs, plus simples et plus abordables.
