La maladie d&rsquo;Alzheimer, une maladie neurod\u00e9g\u00e9n\u00e9rative notoire connue pour son \u00e9rosion progressive de la m\u00e9moire, a \u00e9t\u00e9 un domaine de recherche significatif. Les avanc\u00e9es r\u00e9centes dans l&rsquo;imagerie tau PET, d\u00e9velopp\u00e9es \u00e0 l&rsquo;Universit\u00e9 de Gen\u00e8ve (<a href=\"https:\/\/www.unige.ch\/\">UNIGE<\/a>) et aux H\u00f4pitaux Universitaires de Gen\u00e8ve (<a href=\"https:\/\/www.hug.ch\/\">HUG<\/a>), promettent de remodeler le diagnostic et l&rsquo;intervention pr\u00e9coce de cette maladie d\u00e9bilitante.<\/p>\n\n\n\n<p>La maladie d&rsquo;Alzheimer est caract\u00e9ris\u00e9e par l&rsquo;accumulation de prot\u00e9ines neurotoxiques dans le cerveau, notamment les plaques amylo\u00efdes et les enchev\u00eatrements tau. Comprendre ces accumulations est vital, surtout compte tenu de l&rsquo;\u00e9volution silencieuse de la pathologie de l&rsquo;Alzheimer sur des ann\u00e9es, parfois m\u00eame des d\u00e9cennies. Un diagnostic pr\u00e9coce peut mener \u00e0 des interventions opportunes, qui pourraient ralentir la progression de la maladie.<\/p>\n\n\n\n<p>Actuellement, la tomographie par \u00e9mission de positons (PET) joue un r\u00f4le essentiel dans le diagnostic d\u2019Alzheimer. Valentina Garibotto, qui a dirig\u00e9 cette recherche et occupe des positions \u00e9minentes \u00e0 la fois \u00e0 l&rsquo;UNIGE et aux HUG, pr\u00e9cise : \u00ab&nbsp;Le PET permet de visualiser des processus pathologiques sp\u00e9cifiques dans le cerveau gr\u00e2ce \u00e0 l&rsquo;injection de traceurs radioactifs \u00e0 faible niveau.&nbsp;\u00bb<\/p>\n\n\n\n<p>Alors que les traceurs pour l&rsquo;amylo\u00efde et le m\u00e9tabolisme du glucose existent depuis un certain temps, ils ne fournissent pas une vision compl\u00e8te des complexit\u00e9s d&rsquo;Alzheimer. Le Flortaucipir, un radiotraceur approuv\u00e9 par la FDA d\u00e9velopp\u00e9 en 2020, cible sp\u00e9cifiquement la prot\u00e9ine tau, permettant aux chercheurs de d\u00e9tecter l&rsquo;accumulation de tau et de comprendre sa distribution et ses implications cliniques dans le cerveau.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\">Une nouvelle r\u00e9f\u00e9rence en mati\u00e8re de pr\u00e9diction de la progression de la maladie d&rsquo;Alzheimer<\/h4>\n\n\n\n<p>Dans une \u00e9tude au Centre de M\u00e9moire HUG avec 90 participants, les chercheurs ont trouv\u00e9 que l&rsquo;imagerie tau PET \u00e9tait la plus pr\u00e9dictive du d\u00e9clin cognitif induit par la maladie d&rsquo;Alzheimer. Cecilia Boccalini, une contributrice cl\u00e9 de l&rsquo;\u00e9tude, a soulign\u00e9 sa pr\u00e9cision m\u00eame chez les patients pr\u00e9sentant des sympt\u00f4mes mineurs. Bien que les plaques amylo\u00efdes indiquent la maladie d&rsquo;Alzheimer, c&rsquo;est le tau qui s&rsquo;aligne sur les sympt\u00f4mes cliniques, signalant la progression de la maladie.<\/p>\n\n\n\n<p>Les \u00e9quipes du HUG et de l&rsquo;UNIGE soulignent la promesse d&rsquo;une d\u00e9tection pr\u00e9coce coupl\u00e9e \u00e0 de nouveaux traitements. Comprendre la distribution du tau dans le cerveau est essentiel pour aborder les complexit\u00e9s d\u2019Alzheimer, et int\u00e9grer l&rsquo;imagerie tau PET dans les \u00e9valuations cliniques courantes peut aider \u00e0 cibler des voies th\u00e9rapeutiques personnalis\u00e9es pour les patients, apportant de l&rsquo;espoir aux millions de personnes touch\u00e9es par cette
