Cette avanc\u00e9e a \u00e9t\u00e9 r\u00e9alis\u00e9e en collaboration avec des chercheurs du <a href=\"https:\/\/www.crick.ac.uk\/\">Francis Crick Institute<\/a> et a \u00e9t\u00e9 publi\u00e9e dans la revue scientifique <em>Science Advances<\/em>.<\/p>\n\n\n\n<p>Le nouveau mod\u00e8le de poumon-sur-puce reproduit les alv\u00e9oles humaines, de minuscules sacs d\u2019air responsables des \u00e9changes gazeux et constituant une barri\u00e8re essentielle contre les infections respiratoires telles que la tuberculose. Contrairement aux syst\u00e8mes d\u2019organes-sur-puce pr\u00e9c\u00e9dents, qui combinent g\u00e9n\u00e9ralement des cellules issues de patients et des cellules commerciales, cette nouvelle approche repose enti\u00e8rement sur des cellules d\u00e9riv\u00e9es de cellules souches pluripotentes induites provenant d\u2019un seul donneur. Elle permet ainsi de refl\u00e9ter plus fid\u00e8lement la fonction pulmonaire individuelle et l\u2019\u00e9volution des maladies.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\">Faire progresser une recherche respiratoire personnalis\u00e9e et sans recours \u00e0 l\u2019animal<\/h4>\n\n\n\n<p>\u00c0 partir d\u2019un protocole d\u00e9velopp\u00e9 par l\u2019\u00e9quipe de recherche, des cellules \u00e9pith\u00e9liales alv\u00e9olaires et des cellules endoth\u00e9liales vasculaires ont \u00e9t\u00e9 g\u00e9n\u00e9r\u00e9es \u00e0 partir de cellules souches puis cultiv\u00e9es de part et d\u2019autre d\u2019une fine membrane au sein d\u2019un dispositif de poumon-sur-puce fabriqu\u00e9 par AlveoliX. Le syst\u00e8me est soumis \u00e0 des forces d\u2019\u00e9tirement tridimensionnelles rythmiques simulant les mouvements respiratoires, favorisant la formation de microvillosit\u00e9s et reproduisant \u00e9troitement l\u2019environnement physique des poumons humains.<\/p>\n\n\n\n<p>Afin d\u2019\u00e9tudier la dynamique des infections, des cellules immunitaires issues du m\u00eame donneur ont \u00e9t\u00e9 ajout\u00e9es \u00e0 la puce avant l\u2019introduction de bact\u00e9ries responsables de la tuberculose. Le mod\u00e8le a reproduit des caract\u00e9ristiques cl\u00e9s des premiers stades de l\u2019infection, notamment la formation de regroupements de cellules immunitaires et la d\u00e9gradation progressive de la barri\u00e8re alv\u00e9olaire.<\/p>\n\n\n\n<p>L\u2019\u00e9tude met en \u00e9vidence le r\u00f4le croissant des technologies d\u2019organes-sur-puce pour r\u00e9duire le recours aux mod\u00e8les animaux et permettre une recherche plus pr\u00e9dictive et pertinente pour l\u2019humain. En s\u2019appuyant sur des cellules g\u00e9n\u00e9tiquement identiques, la plateforme ouvre de nouvelles possibilit\u00e9s en m\u00e9decine personnalis\u00e9e, en permettant d\u2019analyser l\u2019influence de contextes g\u00e9n\u00e9tiques sp\u00e9cifiques sur l\u2019\u00e9volution des maladies et la r\u00e9ponse aux traitements.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Des
